Here, we present a protocol for cell transplantation in Nematostella vectensis, a Hexacorallian model. We describe steps for chemotherapy treatment, cell dissociation, and transplantation. We then detail procedures for assessment via fluorescence microscopy and flow cytometry. This technique lays the groundwork for potential coral restoration and conservation applications. For complete details on the use and execution of this protocol, please refer to Talice et al..
