It utilizes high-throughput tools to unravel gene-environment interactions, adaptive biological responses, and molecular pathways impacted by contaminant exposure. While considerable progress has been made in freshwater ecosystems and laboratory species, marine environments and free-ranging wildlife remain understudied. Elasmobranchs, as meso- to apex predators, face significant exposure to bioaccumulative pollutants through trophic transfer. This chronic exposure triggers cellular stress and detoxification responses. Among known molecular markers, Heat Shock Protein 70 (HSP70) and Metallothionein 2 (MT2) are reliable indicators of stress and metal detoxification. However, data on their expression in elasmobranchs are scarce. This study investigated mt2 and hsp70 gene expression in the brain and liver of wild, critically endangered Brazilian Guitarfish (Pseudobatos horkelii) caught by artisanal fisheries in Southeastern Brazil. RT-qPCR and ICP-MS analyses were used to assess gene expression and metal(loid) levels, respectively. Results showed tissue-, sex-, and age-dependent variations in both gene expression and metal accumulation. Correlations between metals, physiological indices and mt and hsp70 gene expressions indicate coordinated responses to metal exposure, reproductive investment, and trade-offs between liver function and body condition, highlighting complex life-stage- and sex-specific metal accumulation and detoxification patterns. The findings support the use of mt and hsp70 for monitoring pollutant exposure and biological responses in threatened elasmobranch species.
