Wang, Mengyao Lai, Qiliang Huang, Zhaobin RNA, Ribosomal, 16S Phylogeny Fatty Acids DNA, Bacterial Bacterial Typing Techniques Base Composition Sequence Analysis, DNA Nucleic Acid Hybridization Geologic Sediments Plant Roots Poaceae Phospholipids Vitamin K 2 Two bacterial strains, designated D2-102 and MBA3-11, were isolated from mangrove surface sediment and cordgrass root, respectively, using the dilution-to-extinction method. The two strains were Gram-stain-negative, coccus-shaped, non-motile, catalase-positive and oxidase-positive. The 16S rRNA genes of the two strains had the highest sequence similarities with WL0086 of 97.6-98.4%, followed by LMO-M01 of 95.5-95.7%. Phylogeny of the 16S rRNA gene placed the two strains into two novel monophyletic branches neighboured by and of the phylum . The digital DNA-DNA hybridization and average nucleotide identity among strain D2-102, strain MBA3-11, WL0086 and LMO-M01 were 19.5-22.8%, and 74.6-79.7%, respectively. The respiratory quinones of the two strains were both menaquinone-7. The major fatty acids (>10%) of the two strains were C, iso-C and anteiso-C. The major polar lipids of the two strains included diphosphatidylglycerol, phosphatidylglycerol and two phospholipids, while phosphatidylethanolamine was only present in strain MBA3-11. Based on the polyphasic taxonomic study, strains D2-102 and MBA3-11 represented two novel species, and thus, sp. nov., and sp. nov. were proposed with type strain D2-102 (=MCCC 1K09973=KCTC 102446) and type strain MBA3-11 (=MCCC 1K09525=KCTC 102445), respectively. Furthermore, based on the phylogeny of the 16S rRNA gene, genomic, phenotypic (including chemotaxonomic) analyses, Ahmad . 2024 should be transferred to the genus as comb. nov. This study expanded the bacterial diversity and ecological distribution of the genus .
