  • The bioproduction of prodigiosin: A comprehensive review on the microbial biosynthesis, regulatory mechanism and cost-effective fermentation strategies. Cui, Yongxiang Xie, Yuheng Salman, Mahwish Adeel, Shahid Kang, Zhen Jiang, Yujia Zhang, Wenming Jiang, Wankui Xin, Fengxue Prodigiosin (PG) is a red tripyrrole pigment belonging to the prodiginine family, which is renowned for its diverse biological activities including anticancer, antibacterial, immunomodulatory and anti-algal effects. Owing to these multifunctional properties, PG has emerged as a promising candidate for applications in pharmaceutical, food preservation and environmental biocontrol etc. Recent advances in PG biosynthesis have deepened our understanding on its biosynthetic pathways and complex regulatory networks. Simultaneously, the discovery of novel wild-type PG producers ranging from marine Pseudoalteromonas sp. to terrestrial Streptomyces sp. has also expanded the prodiginine family, revealing unique structural analogs distinct from canonical PG. Accordingly, this review emphasizes the latest progresses on the structure-function relationship of PG, PG synthetic gene clusters, transcriptional regulation and metabolic engineering. In addition, strategies in PG process fermentation optimization aimed at enhancing PG yields and reducing production costs were also evaluated. These insights will provide a comprehensive theoretical foundation for the rational design of bioprocesses for both PG and its derivatives production, supporting their future development in medical, industrial and ecological areas.
