| contents | Methyl dichloroasterrate alleviates lipopolysaccharide-induced neuroinflammation and depression-like behaviors in mice by upregulating FXR expression. Jia, Yunhui Li, Qinqing Xu, Xinya Kong, Xiangchen Sun, Guangbin Zhang, Zhonghong Zhang, Yifei Li, Li He, Wenbin Liu, Xiangyong Li, Zhipeng Depression is a prevalent mental health disorder closely associated with microglia-induced neuroinflammation. Inhibiting microglial activation can serve as a therapeutic approach by modulating the neuroinflammation cascade. Methyl dichloroasterrate (MD) is a diphenyl ether compound isolated from the fermented metabolites of Aspergillus fungi from marine sources. The biological activity of MD has not been reported previously. In the present study, MD attenuated the lipopolysaccharide (LPS)-induced decrease in sucrose preference in the sucrose preference test (SPT) and diminished the increase in immobility time in the forced swim test (FST) in mice. Furthermore, MD inhibited microglial activation and the expression levels of inflammatory proteins NLRP3 and COX2, ameliorated LPS-induced synaptic damage, and exerted a neuroprotective effect. Additional in-depth research using in vitro and in vivo knockout models showed that MD could increase farnesoid X receptor (FXR) expression, which suppressed the NF-κB pathway and consequently reduced the production of inflammatory mediators. The present study clearly demonstrates that MD elicits anti-neuroinflammatory effects and alleviates depression-like behaviors in mice by upregulating FXR expression. |
