contents High susceptibility of activated lymphocytes to oxidative stress-induced cell death Aníbal E. Vercesi Giovanna R. Degasperi Roger F. Castilho Multidisciplinaria (Ciencias Naturales y Exactas) cell death lymphopenia free radicals immune response spleen lymphocyte The present study provides evidence that activated spleen lymphocytes from Walker 256 tumor bearing rats are moresusceptible than controls to tert-butyl hydroperoxide (t-BOOH)-induced necrotic cell death in vitro. The iron chelatorand antioxidant deferoxamine, the intracellular Ca2+ chelator BAPTA, the L-type Ca2+ channel antagonist nifedipineor the mitochondrial permeability transition inhibitor cyclosporin A, but not the calcineurin inhibitor FK-506, rendercontrol and activated lymphocytes equally resistant to the toxic effects of t-BOOH. Incubation of activated lymphocytesin the presence of t-BOOH resulted in a cyclosporin A-sensitive decrease in mitochondrial membrane potential. Theseresults indicate that the higher cytosolic Ca2+ level in activated lymphocytes increases their susceptibility to oxidativestress-induced cell death in a mechanism involving the participation of mitochondrial permeability transition. 2008 artículo científico 0001-3765 https://www.redalyc.org/articulo.oa?id=32780109 en http://www.redalyc.org/revista.oa?id=327 Anais da Academia Brasileira de Ciências application/pdf Academia Brasileira de Ciências Anais da Academia Brasileira de Ciências (Brasil) Num.1 Vol.80
