The PA containsfour copies of Met-Enkephalin and one copy each of Leu-Enkephalin,heptapeptide and octapeptide. Proenkephalin (aminoacids 1-70)does not contain any opioid peptide sequence andhas been named synenkephalin (Syn). Initial evidences suggestedthat Syn is produced and secreted as an intact moleculeor as part of the precursors in the adult brain and adrenalmedulla, respectively. However, under specific physiologicalconditions, i.e., proliferating immune cells, Syn (8.6 kDa) itis processed to low molecular weight peptides (1.0 kDa). Untilnow there is no evidence of the production or release of a 1.0kDa peptide derived from synenkephalin in the adult rat brain.In this work we studied the postranslational processing ofsynenkephalin in rat striatum, either in whole tissue or in vitrorelease conditions.Male Wistar rats were used throughout this experiment. Therats were sacrificad by decapitation and the striatum removed.Samples were divided into three groups: 1) Thes striatum (n=8)was homogenized and centrifuged; the supernatant was lyophilizedand resuspended in 2 ml water. 2) Striatum was dissectedand sliced in two directions at 90° at 300 mm intervalswith a tissue chopper. The slices were used for the assessmentof the in vitro release of synenkephalin and Met-Enkephalin,using potassium as depolarized stimulus. 3) The synaptosomalfraction was obtained from striatum; samples were depolarizedwith potassium 55 mM, and the perfusates were lyophilizedand resuspended in 2 ml water.
