Velásquez Veterinaria The Murciano-levantina cattlefrom the Spanish South-Eastbelongs to a bovine endangeredbreed under the special protectionof FAO (WWL-DAD:3)*. Theircharacteristics of rusticity, longevity,docility and disease resistancemake interesting the recovery of thebreed. The objective of the presentwork was to determine theefficiency of in vitro bovine embryoproduction using oocytes collectedfrom post mortem ovaries fertilizedwith frozen- thaw sperm obtainedeither from commercial or Murciano-levantina bulls.Cumulus oocyte complexes werematured for 24 h in TCM199 with10% FBS, 2mM L-glutamine, 0.2mM sodium pyruvate, 0.3 μg/mLLH and μg/mL FSH. Oocytespresumed matured were fertilizedwith Percoll-prepared sperm(750.000/mL) in IVF-TALP asdescribed by Parrish et al. (Biol.Reprod 38:1171-1180, 1988).Zygotes (at 18 h) or two-cell stageembryos (at 48 h) were assessedby staining with Hoechst 33342(μg/mL; 20 min) under anepifluorescence microscopy.
