Castaño Medicina RAPD B(a)P Genotoxicity rainbow trout fingerprint ADN In vivo detection of DNA alterations produced by Benzo(a)pyrene using the RAPD technique. The RAPD technique(Random Amplified Polymorphic DNA) is able to detectnonspecific alterations in the DNA of cells with identical genepool, such as in the case of the established cell lines, by bandpattern comparisons of exposed and nonexposed cells. In thepast few years, we have developed a sensitive and highly reproduciblemethodology using RTG-2 cells, an established cell linederived from rainbow trout (Oncorhynchus mykiss). However, itis necessary to test the predictive ability of this assay for in vivostudies. The RTG-2 cell line, as we have previously shown, presents a good genetic similarity with the species from which itderives . Here we show the results of an in vivo study on rainbowtrout individuals sublethally exposed to 69 μg/g bw of benzo(a)pyrene by i.p. injection. The genotoxic effects have beenassessed by band pattern comparison of the DNA that wasobtained from peripheral blood cells taken up at different periodsof time (1 3 months). Due to the fact that that the presenceof polymorphisms interferes with the analysis between the treatedand untreated groups, the comparisons were carried out onthe same individual before and after the treatment.
