contents Gastrocnemius skeletal muscle microvasculature and neuromuscular junction alterations in mice with experimental acute chagas infection Cesare Colasante Maritza Alarcón Elio Moreno Ana Lugo de Yarbuh Veterinaria ischaemia skeletal muscle microvasculature Trypanosoma cruzi acute Chagas´disease A light and transmission electron microscopy study was performedin skeletal muscles (SM) Gastrocnemius (G) from miceexperimentally infected with Trypanosoma cruzi to determinechanges on microvessels (MV) and neuromuscular junction(NMJ) of G. In this study 10 male (mus musculus) (20 g) wereinfected subcutaneally with 1.10 bloodstream trypomastigotesM/DID/Ve/02/DSM strain. Five mice were kept as uninfectedcontrols. The parasites induced a complete paralysis of therear limbs and death while still in the acute Chagas´disease. The histopathology of SM showed inflammatory cell infiltrationby mononuclear and polymorphonuclear leukocytes associatedwith marked parasitism in the muscle fibers of G. Indirect immunofluorescencerevealed interstitial IgG deposit as bandsregularly spaced along the nerve terminals at 40 days postinfection(pi). At this time T. cruzi antigens and intracellularamastigotes nests were also observed. The marked inflammatoryresponse and morphological changes in the SM were confirmedby transmission electron microscopy. Capillary ultrastructurewas seen to be altered, with points of cell cytoplasmdiscontinuity that appear to represent holes in the microvesselwalls. This finding coincided with amastigote nests in myofibers, close contacts between trypomastigotes and endothelialcells and marked thickening of the basement membrane of themuscle vessels. Loss of capillary lumen and a process ofischemia also were observed in the SM of infected mice. Theneuromuscular junction showed degeneration of intramuscularnerve fibers, reduction in the axon caliber, swollen mitochondrial, increase in the actin filaments and microtubules in theaxoplasm, and swelling of the Schwann cells. Increase in thenerve terminal perimeter and most of the synaptic vesicleswere localized near the presynaptic active zones and scarcesin the axoplasm. At this stage of infection the changes findingsin MV and NMJ of G infected with T. cruzi, as well as ischemiaand alterations in the presynaptic membrane densities in theactive zones, shows that the abnormal mice NMJ is associatedwith an activity dependent modulation of the neurotransmission, producing abnormal motor activity and paralysis of therear limbs mice while still in the acute Chagas disease 2006 artículo científico 0798-2259 https://www.redalyc.org/articulo.oa?id=95916605 en http://www.redalyc.org/revista.oa?id=959 Revista Científica application/pdf Universidad del Zulia Revista Científica (República Bolivariana de Venezuela) Num.6 Vol.XVI
