  • <h1><strong><a name="_Toc190636840"></a>Rapid detection and quantification of Antibiotic Resistance Genes (ARGs) and pathogens in wastewater </strong></h1> <p> </p> <p> </p> <p>The protocol allows on-site analysis of resistome in wastewater without the need for samples transport to well-equipped laboratories. The basic protocol outlines DNA extraction procedure and quantitative analysis on the Franklin Biomeme instrument for the following essential (Keenum et al., 2022) for monitoring in wastewater targets: AMR proxies (int1 gene, korB gene), key ARGs (sul1, tetA, ESBL-blaCTX-M-1, ESBL-blaCMY, ESBL -blaSHV, Van) and Acinetobacter baumannii. The Franklin Biomeme qPCR thermocycler is operated from a mobile device running the Android operating system. The Biomeme app can be used to upload the results files to the Biomeme cloud service from where they may be accessed by other authorized users from anywhere. The extended procedure describes the procedure for preparing DNA material for metagenomic analysis on the MinION sequencer (Oxford Nanopore Technologies).</p>
