  • <p><span>We have written a R code, which varies the x-position of the contact point within a window of values A (-0.5 to 0.5 micrometers around the contact point given by the JPK software). For each contact point value, we fit the curve with the model combining elasticity and surface tension (with E and Sigma as free parameters, and initial values of these parameters of 10 kPa, a typical value for cell elasticity (Wu et al. Nat. Methods 2018, doi: 10.1038/s41592-018-0015-1) and 1 nN/micrometer, the tension obtained for oocytes by micropipette aspiration </span><span>(Chaigne et al. 2013, doi: 10.1038/ncb2799)). We calculate the Residual Standard Error (RSE) for each fit. We then retrieve the contact point value corresponding to the minimum RSE in window A and define a new smaller window of values B (-0.2 to 0.2 micrometers around the contact point) and repeat the fitting sequence. In total,  we use 5 steps of “zooming” around the contact point to get the lower value of the RSE and thus accurately determine the contact point. With this method, the contact point value is optimized to achieve the best possible fit, but it is not a free parameter in our expression. To ensure the repeatability of our analysis, the R code used for curve fitting is now accessible here.</span></p>
