contents <p><strong>ABSTRACT</strong></p> <p><br>Checkpoint therapies and CCR5 antagonists reshape immune dynamics but rarely restore com-</p> <p>plete homeostasis. We introduce the FireGate Quad-Axis framework, integrating CCR5 block-</p> <p>ade, PD-L1 modulation, IL-1Ra axis restoration, and broadly neutralizing antibodies (bNAbs)</p> <p>as a modular circuit for immune re-balancing across oncology and neuro-immune repair. IL-1Ra</p> <p>(interleukin-1 receptor antagonist) functions as a stabilizing fourth node, damping inflamma-</p> <p>tory noise while preserving Fc-effector and checkpoint re-activation. In oncology, this axis</p> <p>converts “cold” tumors to “hot,” enabling durable clearance through immune normalization. In</p> <p>neuro-immune contexts such as multiple sclerosis, the same pathway promotes microglial equi-</p> <p>librium and remyelination. Together, these results establish a unified architecture for immune</p> <p>restoration bridging cancer, viral mimicry, and neuroinflammation.</p> <p> </p>
