| contents | <p>Streptococcus suis (Ss) causes the highest economic losses in the pig production industry worldwide and the animal health burden. Ss is responsible for 5% - 20% of mortality in 4-10-week-old age piglets and commonly leads to large consumption (about 50%) of antibiotics in farms. In addition, Ss can cause serious zoonosis such as meningitis, septicemia, or arthritis in humans. Currently, there is no effective vaccine to prevent Ss infection. In this regard, we previously identified the essential genes of Ss causing meningitis and septicemia in piglets, as well as the genome-wide transcriptional profile during infection. The present project aims to acquire knowledge about the functional role of some of these key genes during infection and their potential use for vaccine design. In this project, genes coding for ABC receptors will be studied, particularly gene conservation, amino acid variability, accessibility to antibodies, and antigenicity for pigs, considering a collection of invasive Ss isolates and sera collected from pigs. Then, the protective immune response of the three best-fitting candidates will be validated, separately and in combination, using challenge experiments in standardized animal models. In addition, the specific functions of the genes (encoding nutrient receptors) for bacterial physiology will be identified using isothermal titration calorimetry and growth culture studies, and their involvement in Ss resistance to phagocytosis will be assessed using macrophage culture infection assays combined with Dual RNA-Seq transcriptomic analysis. Altogether, this project provides a better understanding of the streptococcal disease and paves the way to the development of innovative vaccines.</p> |
