contents <p>This methods video is divided into two sections: (1) wet-lab procedures and (2) digital image analysis.</p> <p>In the wet-lab section, we present a resin-based microtechnique that uses an acrylic resin instead of paraffin as the embedding medium. We also describe a resin-optimized trichrome staining protocol that produces a broader color palette than traditional plant tissue stainings. In addition, we demonstrate an immunohistochemistry protocol for resin sections, highlighting practical tips and technical considerations.</p> <p>In the image analysis section, we present a color-based image segmentation workflow implemented in MATLAB R2024a, based on the chromatic differentiation generated by the trichrome staining. The segmented layers are subsequently used for morphological and compositional analyses of tissues and cells in Adobe Photoshop CS6.</p> <p>The resin-based microtechnique provides a more robust alternative to paraffin embedding, enabling high-quality, multicolor, and reproducible plant histological imaging. Combined with systematic image segmentation and quantitative analysis, this workflow enables comprehensive comparisons among tissues and species, paving the way toward “Plant Histolomics.”</p> <p>This video accompanies the manuscript <em>“Plant Microtechnique with Resin: Towards Plant Histolomics”</em>, where methodological details and validation experiments are described in depth.
