contents
Provisional patent specification for USPTO filing. Discloses computational methods for drug repurposing in antibody-mediated inflammatory demyelinating disease (Ab-IDD), encompassing NMOSD and MOGAD, using single-cell RNA sequencing of cerebrospinal fluid from 6 Ab-IDD patients, 3 MS patients, and 2 healthy controls, variational autoencoder-based dimensionality reduction (scVI), Leiden clustering (27 clusters), three-way Wilcoxon differential expression (Ab-IDD vs Normal, MS vs Normal, Ab-IDD vs MS) with ASDC, B cell, and pro-Ab-IDD cluster subset analyses, and LINCS L1000 transcriptomic reversal scoring. Application to GSE194078 across 29 independent Enrichr queries identified 374 NOVEL_ALL candidates (69% of assessed compounds). Top candidates: SAR405838 (MDM2 inhibitor, priority 635,155), R-547 (CDK inhibitor, 600,699), RG-7388 (MDM2 inhibitor, 494,007), AMG-232 (MDM2 inhibitor, 461,017), TAK-733 (MEK1/2 inhibitor, 205,471), MRE-269 (prostacyclin agonist), LXR-623 (LXR agonist). Six MDM2 inhibitors in the top 20 extend cross-disease p53-MDM2 axis findings to five independent tissues. R-547 achieves five-pipeline NOVEL_ALL consistency. The three-way disease comparison design captures Ab-IDD-specific transcriptomic contrast unavailable from binary comparisons. Inventor: Glen Charles Ritschel, Ritschel Research, Tega Cay, SC. Micro-entity filing.
