  • <p class="p1">This dataset contains supplementary files associated with the project on investigating genetic determinants of sexual development and transmission competence in <em>Plasmodium berghei</em>. The deposited materials support analyses of the acetyl-CoA transporter knockout, downstream ACT-responsive genes, and the functional characterisation of candidate regulators including <em>nek3</em>. Files include bulk RNA-seq count matrices, differential expression tables, consensus gene lists, primer tables, single-cell Seurat objects with candidate lineage pseudotime annotations, analysis scripts for bulk RNA-seq, single-cell RNA-seq and structural measurements, and an additional methods document. Together, these supplementary materials provide the primary processed datasets, code, and supporting tables used to examine transcriptional rewiring, gametocyte lineage progression, and candidate gene function following disruption of ACT-associated pathways. The data support the thesis conclusion that ACT acts as an upstream regulator of transcriptional and functional programmes required for male and female gametocyte maturation and malaria parasite transmission.</p>
